Use 2x6 top and bottom plates and offset the stud layout by 8" so that the studs don't align across the plate. So you'd hook the end of the wall and mark one side of the plates at 16, 32, 48, et.
The other side of the wall you'd mark 8, 24, 40, etc. (or cut 8" and mark as usual).

Or cut 3/4" if you want to break on centers.

I staggered my studs midspan from each other. The horizontal members are in-laid for future vertical cedar channel siding (so you wouldn't need that part)

I built walls like that once. It was for an office space inside a factory where the noise level was at something like 100 decibels all the time. It's not that hard to do, I just made up my own routine for doing it. I layed out a stud every 8" on my plates, I think I alternated my layout marks with a line for the first stud, then a line with a slash across it for the next, then just a line, and so on. The line told me to nail the stud flush to the bottom, a line with a slash reminded me to put it flush to the top. The ones I did was with 2x6 plates. Have fun!
 
High "r"

I have been building 2 x 6 walls for several years , but the demand is for the higher r value !!!! And the 2 x 6 plates with the 2 x 4 studs off set helps ,, this next house i plan on using a wet-blown cellulose ( r 3.5 per inch ) and it fills the cracks that fiberglass might not.. And that stops any air flow !!!! Air flow must be stopped first thing to help stop heat loss !!!

Foam is great also , but the cost of foam compared to cellulose ?
 
Is sound an issue?

If so you should really be building two 2x4 walls with a barrier (dead space) in between. The barrier is key in the discontinuation of sound vibration that transfers through framing members. Even a 2x10 wall filled with noise attenuation material would not match two 2x4's with gap.
