Sealing Off Vents On Flatroof With Perlite

Is perlite the way to go for sealing off vents on a flatroof?

My buddy says hes going to be cutting holes in a flatroof for an a/c unit. I dont know the details but he wants to seal it off with perlite. Im guessing the A/C vent comming out of the roof. Is perlite the way to go or should he use some kind of underlayment. I believe its a rubber roof.
